АУТОФАГИЈА У ХЕПАТОЦИТИМА МИШЕВА НА ДИЈЕТИ СА ПОВЕЋАНИМ САДРЖАЈЕМ МАСТИ

Увод: Конзумирање хране са високим садржајем масти заједно са смањеном физичком активношћу доводи до појаве
гојазности и придружених обољења као што је Diabetes mellitus. Diabetes mellitus је хронично, метаболичко обољење које
има све већу преваленцу у свету, а карактерише се хипергликемијом која је настала услед поремећаја секреције инсулина
и/или услед поремећаја у његовом дејству. Тип 2 diabetes настаје као последица резистенције ћелија на инсулин. У типу 2
diabetes-a, аутофагија има значајну улогу јер процес аутофагије обезбеђује ћелији енергију, уклања из ње оштећене
органеле и разне ћелијске агрегате.
Циљ рада: Циљ овог рада био је морфометријска анализа аутофагијских везикула у јетри здравих мишева и мишева
храњених дијетом са високим процентом масти.
Материјал и методе: На електронском микроскопу бројане су аутофагијске везикуле у ћелијама јетре мишева храњених
дијетом са високим садржајем масти (HFD) и мишева храњених стандардном дијетом, а резултат је изражен као апсолутни
број аутофагијских везикула у ћелијама различитих група мишева.
Резултати: Број аутофагијских везикула у хепатоцитима мишева храњених HFD био је статистички значајно већи од броја
аутофагијских везикула у ћелијама животиња храњених стандардном дијетом.
Закључак: Повећање броја аутофагијских везикула у HFD-у указује на постојање промена у одвијању процеса аутофагије у
diabetes mellitus -у.

Introduction: The consumption of high-fat foods coupled with a lack of physical activity are associated with a rise in the prevalence
of obesity and related diseases such as diabetes mellitus. Diabetes mellitus is a metabolic and chronic disorder that has a high
prevalence in the world. The characteristic of diabetes mellitus is hyperglycemia resulting from defects in insulin secretion and/or
insulin action. Type 2 diabetes mellitus is characterized by insulin resistance and failure of pancreatic beta cells to produce insulin.
In type 2 diabetes mellitus, autophagy has a significant role to provide nutrients to maintain cellular energy, removes damaged
organelles and cellular aggregates.
The Aim: The aim of this study was the morphometric analysis of autophagic vesicles in the liver of healthy mice and mice fed with
HFD.
Material and Мethods: Autophagic vesicles in liver cells of different groups of mice (mice fed with standard food and mice fed with
HFD) were counted on electron microscope, and the result is expressed as the absolute number of autophagic vesicles in the cells
of different groups of mice.
Results: The number of autophagic vesicles in hepatocytes of mice fed with HFD was significantly higher than the number of
autophagic vesicles in the cells of mice fed with standard food.
Conclusion: An increase in the number of autophagic vesicles in the HFD indicates the existence of changes in the autophagy
process in diabetes mellitus.
