УТИЦАЈ МАНУКА МЕДА НА DNK ОШТЕЋЕЊА ИЗАЗВАНА ВОДОНИК-ПЕРОКСИДОМ IN VITRO

Увод: Манука мед је монофлорални мед који се добија од цвета дрвета манука (Leptospermum scoparium). Манука мед
испољава већи број биолошких и фармаколошких ефеката, а препознат је као јак антимикробни и антиоксидативни агенс.
Изузетне антиоксидативне особине манука мед поседује захваљујући свом саставу богатом полифенолима и другим
биоактивним једињењима.
Циљ рада: Циљ рада је процена заштитног ефекта манука меда на водоник-пероксидом изазвана ДНК оштећења хуманих
леукоцита периферне крви, применом комет теста.
Материјал и методе: За процену генотоксичности манука меда, ћелије су излагане растворима манука меда у шест
концентрација (25, 50, 100, 250, 500 и 1000 µg/ml). Ради евалуације антигенотоксичног и генопротективног ефекта, узорци
ћелија су третирани у оквиру два протокола, претретмана и посттретмана.
Резултати: Генотоксични ефекти манука меда нису примећени. Eкстракт манука меда je показаo умерено смањење броја
ћелија са ДНК оштећењем у претретману, док је у посттретману испољио већи потенцијал за смањење броја ћелија са ДНК
оштећењем.
Закључак: Манука мед је показао већи антиоксидативни потенцијал на интервентном нивоу. С обзиром да су уочљиви
различити степени смањења оштећења ДНК између претретмана и посттретмана, може се закључити да стимулација
репарацијe ДНК у посттретману несумњиво доприноси значајнијој ефикасности манука меда на интервентном нивоу.

THE INFLUENCE OF MANUKA HONEY ON DNA DAMAGE CAUSED BY HYDROGEN-PEROXIDE IN VITRO

Introduction: Manuka honey is a monofloral honey derived from the flowers of manuka tree (Leptospermum scoparium). Manuka
honey has a number of biological and pharmacological effects, and it is recognized as a strong antimicrobial and antioxidative
agent. The extraordinary antioxidant properties of manuka honey have due to its composition rich in polyphenols and other
bioactive compounds.
The Aim: The objective was to evaluate the protective effect of manuka honey against DNA oxidative damage induced by
hydrogen-peroxide in human peripheral blood leukocytes, using comet assay.
Material and Methods: To evaluate the genotoxic effect of manuka honey, the cells were exposed to solutions of manuka honey in
six concentrations (25, 50, 100, 250, 500 and 1000 μg/ml). For the evaluation of the antigenotoxic and genoprotective effects, the
cell samples were treated within two protocols, pretreatment and post-treatment.
Results: Genotoxic effects of manuka honey have not been observed. Extract of manuka honey showed a modest decrease of DNA
damage in the pretreatment, and in the post-treatment, manuka honey showed higher potential for a reduction of DNA damage.
Conclusion: Manuka honey showed a higher antioxidative potential on an interventional level. Based on the different levels of DNA
damage attenuation that are visible in the pretreatment and post-treatment, we can presume that the stimulation of the DNA
repair mechanisms certainly contributes to the higher efficiency of manuka honey on the interventional level.
